High Protein Diet Exacerbates Azoxymethane-induced Colon Carcinogenesis in Balb/c Mice

Abstract

|
Many epidemiological studies suggested meat, especially red and processed meat, is closely associated with CRC, however, there are very limited evidences on the high protein intake itself causes CRC. We therefore investigated the effect of high protein diet (HPD) on the chemically induced colon carcinogenesis in mice. Twenty female Balb/c mice were randomly divided into 2 experimental groups: normal protein diet (ND, 20% casein, n=10) and high protein diet (HPD, 50% casein, n=10) groups. The mice were initiated with a single intraperitoneal injection of azoxymethane (AOM, 25 mg/kg body weight) and promoted with 3 cycles of 1¡2% dextran sodium sulfate (DSS) in drinking water. Experimental diet was fed to the mice with AOM initiation for 8 weeks. Daily food intake and body weight was lower, however, the weight of large intestine was higher in HPD group than ND group. The length of large intestine was not significantly different between two groups. Colon tumor multiplicity was 279% increased in HPD group compared to that of ND group. Western analysis showed that the expression of COX-2 and iNOS was significantly increased in HPD group. Taken together, this suggests that HPD exacerbates colon tumor formation and increases inflammation-related protein expression compared to that of ND fed mice.
